Myosin II Regulates Growth Cone Actin Dynamics

Abstract

|
Purpose: Myosin II regulates diverse cellular activities, including contraction, migration and adhesion. Rho GTPases also play a similar role in cells. Therefore, it is considered that regulation of myosin II and Rho GTPase activities are closely correlated. In the present study, we aimed at understanding how they regulate growth cone actin dynamics.
Materials and Methods: Neuronal growth cones were investigated in primary cultured hippocampal cells. Alterations in actin dynamics in growth cones were monitored following treatment with blebbistatin (BBS), a specific myosin II ATPase inhibitor. Myosin II phosphorylation was analyzed by immunoblotting and immunocytochemistry.
Results: Hippocampal growth cones showed disordered actin structures 30 min following treatment with BBS. Removal of BBS restored their original shapes, indicating that BBS-induced alterations in actin dynamics are reversible. Stimulation with NGF did not induce profound changes like BBS, but myosin II phosphorylation was detected by immunlblotting and immunocytochemistry.
Conclusion: It is suggested that myosin II regulates growth cone actin dynamics through Rho GTPases.
